The Effects of Phenothiazine Derivatives on the Osmotic Fragility and the Potassium Transport of Rabbit Red Cells

Abstract
|
|
|
The effects of phenothiazine derivatives such as chlorpromazine, promazine, perphenazine and prochlorperazine on the permeability of rabbit red cells to potassium were studied by analyzing dose-response, time-response curves and the changes of osmotic fragility in the saline medium of concentration ranging between 0. 3 and 0. 9%. The results were as follows.
1. Phenothiazine derivatives such as chlorpromazine, promazine, perphenazine and prochlorperazine induced a loss of K+ from rabbit red cells in vitro in the following order: chlorpromazine> perphenazine> prochlorperazine> promazine.
2. Osmotic fragility of rabbit red cells was increased by the administration of phenothiazine derivatives such as chlorpromazine, promazine, perphenazine and prochlorperazine in the same order as they did in the loss of K+.
3. Blood plasma, dextrose and albanin protected red cells against the loss of K+ and the increase of osmotic fragility induced by chlorpromazine, promazine, perphenazine, and prochlorperazine, but EDTA did not protect.
